Echoes of Immunity Past: Decoding the Ubiquitous, Highly-individualized, and Surprisingly Stable World of Autoantibodies

There is no such thing as an immunologic twin: Despite identical genomes, monozygotic twins develop remarkably different antiviral and autoantibody profiles that remain stable for decades (HuProt Microarrays, VirScan PhIP-Seq, HuScan PhIP-Seq,).

Syngeneic mice contain diverse serum antibody profiles: Environmentally controlled inbred mice still develop distinctive individual autoantibody signatures, challenging the concept of biological replicates in preclinical research (HuProt Microarrays, MouseScan PhIP-Seq).

Disease signals stand out from individual noise: While the majority of autoantibodies remain unique to each individual regardless of health status, disease-associated signatures emerge in inflammatory bowel disease, Sjogren’s, dementia, and more (HuProt Microarrays, VirScan PhIP-Seq, HuScan PhIP-Seq).

This webcast will highlight novel technologies that enable the detection of autoantibodies by screening human serum samples against more than 80% of the human proteome. The presence of autoantibodies may indicate immune recognition of melanoma cells and correlate with disease progression, response to therapy and occurrence of treatment-related side effects. While autoantibody panels could enhance diagnostic accuracy and disease monitoring, challenges remain in standardising these biomarkers due to variability in their data analysis pipeline, immune response variability and specificity concerns. Future research integrating autoantibody profiles with other molecular and imaging biomarkers may improve melanoma detection and patient outcomes.

In this GEN webinar, our speaker, Eric Johansen, PhD, will demonstrate the utility of virion-displayed membrane proteins for screening antibody biologics. He will present the VirD membrane protein array technology, which displays membrane proteins in the membrane envelope of herpes simplex virus-1 (HSV-1) virions. The technology is designed for high-throughput biochemical analysis of membrane proteins. Membrane protein expression occurs during virus replication, ensuring that the integrated proteins undergo endogenous folding and post-translational modifications in their native and active conformations. He will also share the results of testing various commercial and clinical antibodies.

While autoantibodies have traditionally been suspected in disease areas like multiple sclerosis and rheumatoid arthritis, there is a growing body of evidence that autoantibodies are involved in a diverse range of disease areas such as cancer (immunotherapy outcomes and putative therapeutics), obesity, psychiatric disorders, neurodegenerative diseases, and may others. Oftentimes these autoantibodies are formed in response to viral infections. It is increasingly clear that each individual’s autoantibody repertoire is incredibly unique, complex, and yet accessible and understandable via CDI Labs’ HuProt™ Microarray and VirScan®, HuScan®, MouseScan™ PhIP-Seq (phage immunoprecipitation sequencing) assays.
